2FA or Yubikey with a vault

Is there any option for utilizing a Yubikey or some form of 2FA with a vault?

No.

It is on our longterm roadmap and we already applied some changes to later integrate it, but as of now, 2FA is not supported.

An interesting read is the discussion on our issue tracker about 2FA: https://github.com/cryptomator/cryptomator/issues/367

Yubikeys are only used for authentication and not for encryption (correct me if I am wrong).
Those are two completely seperate things.

You are not wrong, but the question pertains to using a Yubikey to access a vault, nothing to do with encryption.

Yubikey can do not just only authentication, Yubikey supports HMAC-SHA1, PIV and OpenPGP, HMAC-SHA1 can derive encryption key, PIV and OpenPGP can used for encryption.
