Perhaps something that was asked before but I can’t find any reference to it.
Say I create a Vault on my local NAS (Samba share), can I mount the very same vault from multiple LAN devices? e.g. does Cryptomator account for multiaccess?

Yes, thats possible
Cryptomator itself not, but your OS should.
Means if one files is blocked because you have opened it from device A, and you open it with device B, then you should receive the same message as if you would do the same with an unencrypted file.
I am speaking in theory because I do not have a vault on a NAS. But you can just try and find out
